What is Naloxone?
Active Ingredients: Naloxone HCl
Naloxone HCl Injection is used to treat a suspected opioid overdose or reverse opioid depression due to opioids, nalbuphine, pentazocine, or dextropropoxyphene. It is also administered to treat newborns with respiratory or CNS depression that is caused by analgesic administration to the mother.
Naloxone is classified as a centrally-acting opioid antagonist. It works by blocking the effects of opiates in the brain.

What is Naloxone used for?
It is used to block or reverse the effects of opioid medications.
How is Naloxone Injection used?
This medication is manufactured as a solution, which is intended to be administered by intravenous, intramuscular or subcutaneous injection or by IV infusion.
What dose should be given?
Adults
- For opioid overdose, the usual dose for adults is 0.4-2mg, given IV/IM or SC. If needed, repeat doses at 2-3 minute intervals. After a total dose of 10mg, if there is no improvement, the diagnosis should be reevaluated.
- To reverse opioid sedation, the usual initial dose for adults is 0.1-0.2mg, given at 2-3 minute intervals. Smaller doses are typically sufficient for the partial reversal of opioid depression after surgery.
Children
- For opioid overdose in children, the usual dose is 0.01mg/kg, given IV. The dose may be raised to 0.1mg/kg if needed.
- To reverse opioid sedation in children, the usual dose is 0.005-0.01mg/kg, given IV. Repeat doses at 2-3 minute intervals, as needed.
Neonates
- For opioid overdose in neonates, the usual dose is 0.01mg/kg, given IV/IM/SC. Repeat doses at 2-3 minute intervals, as needed.
- To reverse opioid sedation in neonates, the usual dose is 0.01mg/kg, given IV/IM/SC. Repeat doses at 2-3 minute intervals, as needed.

Side Effects
As with all pharmaceuticals, some unwanted effects can occur from the use of Naloxone HCl Injection.
Naloxone may cause sudden opioid withdrawal symptoms, such as:
- gastrointestinal effects (nausea, vomiting, diarrhea, abdominal pain)
- sweating, fever, or body aches
- tremors, shivering, tachycardia
- runny nose
- restlessness or irritability
In infants younger than 4 weeks old, withdrawal symptoms may be life-threatening if not treated. Symptoms may include crying, stiffness, or seizures.
