What is Domperidone?
Active Ingredients: Domperidone
Domperidone Tablets are an antiemetic drug used for the short-term treatment of nausea and vomiting. This medication is also prescribed to treat stomach pain in patients during palliative care. Domperidone may be used to increase breast milk production in women having trouble breastfeeding.
Domperidone is sometimes used to relieve gastrointestinal symptoms, such as indigestion or dyspepsia. It can be used as a peristaltic stimulant in order to treat symptoms associated with (gastroparesis) delayed gastric emptying, including abdominal distention, epigastric pain, and sense of fullness.
Domperidone is a dopamine antagonist. It works by blocking the action of the chemical substance that triggers the feeling of nausea and vomiting in the brain.
Domperidone Tablets are produced with a dosage of 10mg. The tablets are available for distribution as a box of 10 tablets.

Uses
What is Domperidone used for?
It is used to relieve nausea and vomiting. Domperidone is sometimes used to treat symptoms, including indigestion, upset stomach, epigastric pain, and abdominal swelling, that are associated with gastrointestinal conditions, such as gastroesophageal reflux disease (GERD), gastroparesis, and functional dyspepsia.
How should Domperidone Tablets be used?
This medication is intended to be taken orally. It is recommended to take the tablet with a full glass of water. It is most effective when taken 15-30 minutes before a meal. Though the medication can be taken after eating, it may take a longer time to work.
What dose should be taken?
Adult Dosing Dosage may vary based on different medical indications:
- For nausea and vomiting, the usual dose is 10mg, taken 20 to 30 minutes before meals. The dose may be repeated up to 3 times per day. The maximum dose is 30mg per day.
- For gastrointestinal pain (palliative care), the usual dose is 10mg taken 3 times per day.
Pediatric Dosing Recommended dosage for children may vary based on different medical conditions:
- For nausea and vomiting in children over 12 years old with a body weight of more than 35kg, the usual dose is 10mg, taken 20-30 minutes before meals. The dose may be repeated up to 3 times per day. The maximum dose is 30mg per day.
- For GERD in children from 1 month to 18 years old (off-label), the usual dose is 25mcg/kg, taken three times per day. The maximum dosage is 10mg per dose. If the response to the drug is insufficient, a dose of 400mcg/kg can be considered, with a maximum dosage of 20mg per dose. It is advised to stop treatment at occasional intervals in order to assess the recurrence of symptoms. If symptoms return, treatment may be restarted. Therapy should be discontinued if the response is insufficient at high doses.
The dosage is based on medical condition, response to treatment, age, and weight. Refer to a doctor or pharmacist for guidelines on dosage. Do not exceed what they advise.
What happens if a dose of Domperidone Tablets is missed?
If a dose is missed, it can be taken as soon as it is remembered. If it is nearly time for the next dose, the missed dose should be skipped. Never take 2 doses at the same time.
Who can use Domperidone?
Domperidone Tablets can be given to adults and children, but caution is advised for specific groups of patients.
Pregnant This drug is not recommended for use in pregnant women and should only be used if the benefits outweigh the risks. Limited data and studies are available to assess the risk of fetal harm.
Breastfeeding Domperidone is not recommended for use by breastfeeding women and should only be used if there is a clear need. A small amount of the drug passes into milk, and there is a risk of cardiac side effects in the infant. Though Domperidone is occasionally used to increase milk supply, there have not been enough studies to assess safety guidelines for this use. It is advised to express caution when giving this medication to nursing infants with QTc prolongation risk, unstable GI function, or any serious illness. Caution should also be used for neonates, premature infants, and infants with low birth weight.
Children This medication is considered safe for use in adolescents and children over 12 years old. It is not recommended for children under 12 years old. Though Domperidone is used in younger children and infants, the tablet formulation is not the preferred route of administration. It is important to note that there is an increased risk of neurological side effects in younger children, as the blood-brain barrier and metabolic functions are not fully developed.
Geriatric Domperidone is considered safe for use in elderly patients up to 60 years old. Caution should be used for patients over 60 years old, as there is an increased risk of ventricular arrhythmias or sudden cardiac death.
How should Domperidone Tablets be stored?
It is recommended to keep this medication in its original packaging until use. The tablets should be kept in a dark location at room temperature (20-25°C). Keep all medications out of the reach of children or pets.
Any medication that is past its expiry date should be discarded in accordance with local regulations.
Other warnings
For patients with severe renal impairment, dose adjustments should be considered. It may be necessary to reduce dosage to 1 or 2 times per day, depending on the severity of illness. It should be noted that a single dose per day is unlikely to need adjustment.
Domperidone is associated with elevated levels of prolactin due to its ability to block the D2 receptor.
The use of Domperidone may increase the risk of cardiac arrhythmia or sudden cardiac death. It should be used cautiously in patients older than 60 years old, who have pre-existing risk factors for QT prolongation, or who are taking QT-prolonging drugs or CYP3A4 inhibitors. It is advised to avoid use in patients with cardiac disease, cardiac conduction interval prolongation, severe electrolyte imbalances, and moderate/severe renal impairment.
This drug should be used at the lowest effective dosage and for the shortest possible time in order to reduce the risk of cardiac events. It is recommended to advise patients to consult emergency services in the event of arrhythmia.
Side Effects
As with all pharmaceuticals, some unwanted effects can occur from the use of Domperidone Tablets.
Common side effects include, but may not be limited to:
- dry mouth
- headache
- hives or itching
- hot flashes
- anxiousness
- drowsiness
- diarrhea
- rash or itchiness
- general weakness
- tender or painful breasts
Serious side effects may include:
- dizziness, lightheadedness, fainting, or disorientation
- difficulty speaking
- seizures
- signs of an allergic reaction
- bradycardia or other changes in heart rate

Precautions
Do NOT use Domperidone Tablets if:
- You are allergic to any of the ingredients.
- You have breast cancer (prolactinoma).
- You have moderate to severe hepatic impairment.
- You have torsade de pointes.
- You have severe heart disease.
- You have a history of allergies or allergic reactions.
- You have GI bleeding, obstruction, or perforation.
Before treatment, consult your doctor regarding any medications you are taking to address potential drug interactions.Domperidone is known to interact with medications such as antidepressants (citalopram, escitalopram), antipsychotics (haloperidol, pimozide, sertindole), antibiotics (erythromycin, clarithromycin, telithromycin, moxifloxacin, pentamidine), antifungals (ketoconazole, fluconazole, voriconazole), antimalarials (halofantrine), HIV medications (protease inhibitors), drugs for hypertension or heart problems (amiodarone, dronedarone, quinidine, disopyramide, dofetilide, sotalol, diltiazem, verapamil), or anticancer medications (toremifene, vandetanib, vincamine). This is not a complete list.
It is recommended to avoid grapefruit products while being treated with domperidone. Grapefruit has been associated with increased plasma levels of domperidone.
This medication may not be suitable for people with certain conditions, so it is important to consult with a doctor if you have any health conditions. Special considerations, additional monitoring, or adjusted doses may be necessary for conditions, such as:
- body weight of less than 35 kg
- electrolyte imbalances
- kidney or liver impairment
- breastfeeding or pregnancy
- galactosemia, glucose-galactose malabsorption syndrome, hereditary fructose intolerance, or lactose intolerance
- history of torsade de pointes
Some formulations of domperidone may contain inactive ingredients such as lactose or sorbitol. It is advised to consult the packaging insert for a full list of ingredients.
Dizziness and drowsiness have been reported as side effects of this medicine. It is advised to avoid driving or operating machinery until it is clear how the medication affects you.
This medication should only be used for the shortest duration possible. If symptoms have not subsided within 1 week, contact a healthcare professional.

Safety of domperidone in treating nausea associated with dihydroergotamine infusion and headache
The main goal of this study was to determine the safety of domperidone in the treatment of nausea associated with dihydroergotamine (DHE) infusion and headache.
During the study, 103 consecutive patients, using domperidone, were admitted for IV DHE by infusion. Most admissions were for the treatment of chronic migraine, either with aura (n = 53) or without aura (n = 46). Domperidone was given in 85 out of 103 cases and was well-tolerated at doses of up to 80mg per day. This drug in combination with other antiemetics is effective in treating nausea.
The conclusion of the study is that domperidone is safe in the treatment of nausea associated with inpatient DHE infusion and headache. Patients with headaches undergoing DHE infusion can also use domperidone to reduce the nausea.
